研究内容

動物細胞の細胞分裂は分裂面の膜直下に形成されるアクチン繊維とミオシンからなる収縮環の収縮によって引き起こされる。現在、分裂酵母や細胞性粘菌、ショウジョウバエなどを用いて、収縮環が形成される位置決定機構や、収縮環の形成・収縮・消失の分子機構が世界中で研究されている。我々は繊毛虫テトラヒメナを用いて、世界に先駆けて分裂面の位置決定に関わる蛋白質(p85)を同定し、p85がCa2+/カルモデュリン(CaM)によって調節されていることを発見した。我々は現在、収縮環形成機構に関してさらに研究を進めている。

この過程でリボソームでの蛋白質翻訳で重要な働きをしているペプチド伸長因子1α(EF-1α)が分裂溝でアクチン繊維束形成に関わっていることを発見した。さらにテトラヒメナのクエン酸合成酵素も多機能性を持つことを発見した。このような多機能蛋白質の存在が、生物が限られた遺伝子を有効に利用するために必要不可欠であったのではないかと考え、多機能蛋白質の生物学的意義に関して研究を進めている。

(1) 分裂シグナルの実体の解明

細胞分裂の分裂溝の位置は分裂シグナルによって決められていると考えられている。分裂溝の位置決定に異常を持つ突然変異株cdaA1を用いて、分裂位置決定に関与する蛋白質(p85)を同定した。p85はCa2+結合蛋白質であるCaMと結合し、両者は分裂溝に共局在した。また、CaMの阻害剤であるW7はp85とCaMの結合、両者の分裂溝への局在、分裂の進行を阻害した。これらは分裂シグナルの実体がCa2+/CaMであることを示唆している。さらにp85はCa2+/CaMに依存してアクチン単量体と結合し、分裂面にアクチン分子を集積させる働きがあった。その結果、分裂面でアクチン濃度が上昇し、アクチン重合が速やかに進み収縮環が形成されるものと予想される。分裂溝に局在してアクチン重合を促進しているプロフィリンが収縮環形成過程にどのように関わっているかを調べるため、プロフィリンと相互作用するフォルミンファミリーの蛋白質、BNI1とBNI2の性状と機能を調べている。その結果、BNI1はプロフィリンと結合し、分裂溝に局在することが明らかになった。

(2) 収縮環の形成機構の解明

収縮環の形成・収縮・消失を調節するアクチン結合蛋白質の機能を明かにするためテトラヒメナより2種類のアクチン結合蛋白質を単離精製した。その1つであるEF-1αはアクチン繊維を束ねる活性を持ち、その活性はCa2+/CaMによって制御されており、EF-1αとCaMは分裂溝に共局在していた。もう1つはフィンブリンでアクチン繊維を束ね、分裂溝に局在していた。細胞分裂におけるこれらの蛋白質の機能を遺伝子破壊や遺伝子改編の技術を用いて調べている。

(3) 収縮環の収縮機構の解明

収縮環の収縮はアクチン繊維とミオシンの間に生じる滑り運動によるものと考えられているが、ミオシンが収縮環内でどのような状態で存在するかは明らかになっていない。そこでテトラヒメナよりミオシンIIに類似したミオシンを精製し、さらにテトラヒメナから13種類のミオシン遺伝子のクローニングに成功した。それら13種類のミオシンの性状と機能を網羅的に調べるプロジェクトをスタートさせ、口部装置からの食胞形成と細胞肛門からの排出に係るユニークなミオシン13を発見した。今後、収縮環内でアクチンと相互作用するミオシンの実体を明らかにしたいと考えている。

(4) 多機能蛋白質の研究

テトラヒメナから、全く異なる2つの機能を持つ多機能蛋白質、クエン酸合成酵素/14nm繊維蛋白質とEF-1α を発見した。クエン酸合成酵素はミトコンドリアでは酵素として機能し、細胞質では細胞骨格として機能しており、これらの機能はリン酸化脱リン酸化によって制御されていた。ゲノムプロジェクトの結果、遺伝子の数はヒトでは約35,000、ハエでは約18,000であリ、かなり少ないことが解った。限られた遺伝子を有効に利用するためには gene sharing が必要であり、その結果が多機能蛋白質の存在であると考える。多機能蛋白質が数多く存在し、普遍的なものであるかどうかを検証し、その生物学的意義を解明したい。

(5) 高分子ポリフェノールの研究

我々はテトラヒメナとローダミン123を用いてミトコンドリア活性化因子の探索法を開発した。この方法で、ウーロン茶や紅茶にミトコンドリア活性化因子(MAF:mitochondrial activation factors)があることを発見し、その精製に成功した。MAFはテトラヒメナのATP産生量、酸素消費量、繊毛運動速度を増加させた。また、MAFはマウス精子のミトコンドリア膜電位と精子鞭毛運動の速度も増加させ、2型糖尿病モデルマウスの血糖値を下げ、肝脂肪を減少させるという多様な効果を持つことが分かった。メタボやサルコペニアを予防する発酵茶から抽出する高分子ポリフェノールMAF
